A first genetic linkage map of the blue mussel Mytilus edulis ArchiMer
Lallias, Delphine; Hecquet, Céline; Boudry, Pierre; Lapegue, Sylvie; Beaumont, Andy.
The blue mussel, Mytilus edulis, is widely distributed in the northern hemisphere, from the White Sea to the Atlantic coast of southern France, and is an economically important species (1 million tons per year, FAO, 2002). At present however, little domestication has been achieved with bivalves, in contrast to agricultural crops, livestock and horticultural species like tomatoes, where selection and production is entirely controlled by man. For bivalves, despite their economic importance, no lines with selected characteristics have been taken to the production scale. There is therefore a need to make up for lost time. Our interest in genetic mapping sprang from some results showing a genetic component to some economically important traits like growth and...

A first genetic linkage map of the European flat oyster, Ostrea edulis : towards the identification of Bonamia resistance QTLs ArchiMer
Lallias, Delphine; Lapegue, Sylvie; Haley, Chris; Heurtebise, Serge; Boudry, Pierre; Beaumont, Andy.
The flat oyster Ostrea edulis is an endemic European oyster species, found on both Atlantic and Mediterranean coasts. Its aquacultural production has dramatically decreased due to two successive diseases caused by the intracellular parasites Marteilia refringens and Bonamia ostreae. Since 1985, Ifremer has undertaken a breeding program to produce oyster families which are tolerant to Bonamia. In this context, the next step is to identify QTLs for resistance to this parasite. A genetic map is therefore being established as a basis for QTL work. The reference family is an F2 family produced by single pair mating of two oysters from the same F1 family. The F1 family was itself produced from a bi-parental cross between a wild oyster and an individual from the...

A preliminary genetic linkage map for the European flat oyster Ostrea eduli ArchiMer
Lallias, Delphine; Lapegue, Sylvie; Halley, Chris; Boudry, Pierre; Beaumont, Andy.
The European flat oyster, Ostrea edulis, occurs naturally from Norway to Morocco in the North-Eastern Atlantic and throughout the Mediterranean. The oyster has been a harvested species for at least 6000 years. However, it successively suffered from over harvesting and then from two protozoan diseases during the 1960s and its production has drastically decreased in Europe from that time. Following two generations of selection for tolerance to one of these diseases (bonamiosis), our aim is to optimize this selection with the assistance of molecular markers, that is to say to implement marker-assisted selection. This will enhance the efficiency of selection by reducing the generation time, as the selection on sites cannot be made before the oysters are...

A preliminary genetic linkage map for the European flat oyster Ostrea edulis L. ArchiMer
Lallias, Delphine; Lapegue, Sylvie; Harley, Crhis; Boudry, Pierre; Heurtebise, Serge; Beaumont, Andy.
The flat oyster Ostrea edulis is the species endemic to European coasts, both Atlantic and Mediterranean. It has been exploited since Roman times, but its aquaculture production decreased from around 20000 tons in the 1950's to 1500 tons today because of two successive diseases due to the intracellular parasites Marteilia refringens and Bonamia ostreae. Since 1985, Ifremer has been undertaking a breeding program to produce families of oysters tolerant to Bonamia. In this context, a further step would be to identify QTLs for resistance to this parasite. Therefore, as a first step a genetic map is now being built. The establishment of a genetic linkage map will provide a foundation for the mapping of QTLs, with the ultimate objective to implement...

Lapegue, Sylvie.
In the framework of the French diploma "Habilitation à Diriger des Recherches", Sylvie Lapègue's synthesis of research includes ten years (1998-2007) of work in bivalve genetics, and mainly cupped and flat oysters. This research has been developed in (1) flat oyster population genetics at the European level and cupped oyster populations and species genetics at a global scale, (2) the characterization of a cytogenetic particularity observed in oysters that corresponds to a high rate of aneuploidy in somatic cells, and (3) more recently in the use of molecular markers for the construction or improvement of genetic maps, allowing (4) the identification of areas of the genome related to the resistance to summer mortality in the Pacific oyster and the...

IDENTIFICACIÓN DE QTLs ASOCIADOS A CARACTERES DE ARQUITECTURA VEGETAL EN YUCA (Manihot esculenta) Acta biol.Colomb.
MORA MORENO,Ruben Eduardo; SOTO,Johana Carolina; LÓPEZ,Camilo.

(Palmira)
Morillo C,Ana Cruz; Morillo C,Yacenia; Ceballos L,Hernán.

Linkage disequilibrium, genetic association mapping and gene localization in crop plants Genet. Mol. Biol.
Sorkheh,Karim; Malysheva-Otto,Lyudmyla V.; Wirthensohn,Michelle G.; Tarkesh-Esfahani,Saeed; Martínez-Gómez,Pedro.
DNA-based molecular markers have been extensively utilized for a variety of studies in both plant and animal systems. One of the major uses of these markers is the construction of genome-wide molecular maps and the genetic analysis of simple and complex traits. However, these studies are generally based on linkage analysis in mapping populations, thus placing serious limitations in using molecular markers for genetic analysis in a variety of plant populations. Therefore, alternative approach has been suggested, linkage disequilibrium-based association analysis which detects and locates quantitative trait loci (QTL) by the strength of the correlation between a trait and a marker. Although association analysis has already been used for studies on genetics of...

Overwiew on molecular tools in bivalve shellfish aquaculture ArchiMer
Boudry, Pierre; Saavedra, Carlos.
Dr. Boudry presented an overview of the current development of molecular tools in shellfish bivalves, focussing on the recent development in oysters, species to which the most efforts are dedicated. This review was prepared together with Dr. Saavedra (Mollusc Research Group, Torre de la Sal, Spain) within the Fish and Shellfish group of the EU Network of Excellence "Marine Genomics Europe". Searches in DNA sequence databank ("GenBank") show that 4% are microsatellite sequences, 23% are mitochondrial sequences and the remaining 73 % other nuclear sequences. Among these, 75% concern oyster (Crassostrea) species, 15% mussel (Mytilus) species and 10% scallop (Argopecten) species. Some of these sequences were targeted as candidate genes related to known...

QTL mapping for resistance to bonamiosis in the European flat oyster Ostrea edulis ArchiMer
Lallias, Delphine; Beaumont, Andy; Haley, Chris; Heurtebise, Serge; Boudry, Pierre; Lapegue, Sylvie.
A breeding programme has been undertaken by Ifremer to produce selected Ostrea edulis families which are resistant to Bonamia ostreae, a parasite which is the most significant constraint on aquaculture of this oyster. Using such families, we aimed to identify Quantitative Trait Loci (QTLs) of resistance/susceptibility to bonamiosis. We used an F2 family derived from a bi-parental cross between a wild oyster and an individual from one of the selected oyster families. A total of 550 F2 individuals from the family cohabited with wild oysters that were injected with the parasite. Mortality was checked daily over six months and heart smears were used on moribund or dying oysters to detect the presence and level of infection of the parasite. Ninety two F2...

Quantitative trait locus mapping associated with earliness and fruit weight in tomato Scientia Agricola
Hernández-Bautista,Aurelio; Lobato-Ortiz,Ricardo; García-Zavala,José Jesús; López-Fortoso,Fernando; Cruz-Izquierdo,Serafín; Chávez-Servia,José Luis; Cadeza-Espinosa,Marisol.
ABSTRACT The flowering time is regarded as an important factor that affects yield in various crops. In order to understand how the molecular basis controlling main components of earliness in tomato (Solanum lycopersicum L.), and to deduce whether the correlation between fruit weight, days to flowering and seed weight, is caused by pleiotropic effects or genetic linkage, a QTLs analysis was carried out using an F2 interspecific population derived from the cross of S. lycopersicum and S. pimpinellifolium. The analysis revealed that most of the components related to earliness were independent due to the absence of phenotypic correlation and lack of co-localization of their QTLs. QTLs affecting the flowering time showed considerable variation over time in...

The use of SNPs in characterizing oysters genomes and their resistance to pathogens ArchiMer
Lapegue, Sylvie; Heurtebise, Serge; Harrang, Estelle; Morga, Benjamin; Flahauw, Emilie; Sauvage, Christopher; Boudry, Pierre.
During the last five years, a great effort has been made to develop genomic tools in oyster species all over the world. In Europe, the Pacific cupped oyster, Crassostrea gigas, and the European flat oyster, Ostrea edulis received a special attention as they are the key species for local aquaculture. In particular, the Pacific oyster is a marine bivalve of major economical and ecological importance worldwide. Better knowledge of its physiology and its genetics contributes to optimize its aquaculture production using selective breeding or to understand its invasive behavior in some coastal areas. Current international developments of genomic resources include EST databases, BAC libraries, genome mapping and sequencing. The Genoscope (Evry, France) and the...
